Research Interests

Our goal is to understand the role of spatial chromatin organization in the regulation of gene expression in mammalian cells. We are particularly interested in defining how epigenetic modifications affect chromatin structure and genome function in "normal" and cancer cells. We are characterizing the molecular mechanisms involved in regulating the three-dimensional chromatin architecture and expression of Hox genes in leukemia cell models.

HoxA CTCF Sites in Motion

The conformational changes associated with HoxA 5' end gene regulation throughout the THP-1 differentiation time course is shown. The CTCF sites are represented by red spheres. Yellow spheres indicate H3K27me2/3 signals above a threshold with a cutoff = 0.8. Pauses between transitions identify the models at each of the five time points. Models between the time points are interpolated to produce a smooth transition. (Rousseau et al.)
