My project consists of understanding how the 3D chromatin organization affects the regulation of the HoxA genes during limb development using mouse model.
HoxA genes are developmental genes and the strict control of their expression is crucial for proper embryonic development. These genes constitute a good model to study gene regulation during development.
For my study, I optimized several "Chromosome Conformation Capture" techniques to analyze the chromatin architecture in tissues. I used genetic and genomic approach to identify limb-specific regulatory elements controlling HoxA gene expression.
This study allowed us to identify several regulatory elements regulating the HoxA gene during limb development.
I am currently studying the mechanisms controlling regulatory element specificity that allows a precise gene regulation during development.
